What device string are you typing into Xastir?  "/dev/ttyS0" or "/dev/USB0" perhaps?

What hardware port are you actually using to connect to the TNC?  A true RS-232 serial port, or a USB->Serial cable?

That will get the discussion started.  Once we figure out the above, we next need to look at ownership/permissions for the port you're using.
